Enyimba, who were drawn bye in the first preliminary round, will now tackle Etoile de Filante from Burkina Faso in the next round of the competition.
Enyimba, who finished third in the NPFL last season, will travel to Burkina Faso on September 13 for the first leg, before the return leg a week later in Nigeria.
Etoile de Filante reached this stage of the competition after they eliminated Hafia of Guinea.
The first leg in Guinea ended in a 1-1 draw, before the return leg in Burkina Faso which finished goalless for Etoile de Filante to scrape through on the away goals rule.
